In Sekiro: Shadows die twice, some enemies with a lot of health, minor bosses, or difficult locations to access drop prayer beads. Getting most beads is optional, and some are mandatory. While It is possible to beat the game by only getting the necessary beads, more beads are better. When you have four of them, you can craft a necklace at a sculptor’s idol to increase vitality and posture. I completed the immortal severance run with only four necklaces, but there are 40 total beads, thus, 10 total necklaces. While many beads come from minor bosses, others can be found in various locations on the ground, so be sure to pick up everything you possibly can. On the playthroughs after your first one, you get a second chance to get the beads you may have missed.

Like I mentioned before, I finished my first playthrough With four necklaces (16 beads). I found this to be a sufficient amount, even though I only got some of the easy beads. To list some bosses that drop beads: General Naomori Kawarada, The Chained Ogre, and Tenzen Yamauchi, the raging bull, and so on. These are all minor bosses, and throughout my playthrough I began to predict if a boss would drop beads, or a battle memory.
